及び研究内容の紹介 |
カリキュラムに加え、早期から基礎医学研究を実践するプログラムを希望することができます。本プログラムでは、1,2 年次は希望者を対象とした研究紹介や基礎医学研究の体験実習に参加し、3 年次以降は少人数プログラムとなり本格的な研究が開始します。私も昨年度から研究を開始し、3 月の第120 回日本解剖学会総会・第92 回日本生理学会大会・合同大会と連動して行われたMD プログラム全国リトリートや、6 月の第99 次日本法医学会学術全国集会Student Poster Forum などでポスター発表をさせて頂きました。
しかし、今年に入ってからは臨床講義の開始に加え、個人的な理由も重なり、恥ずかしながら研究は進められていない状況です。研究内容は「Effect ofheat on the immune system in organotypic brain slice cultures」と題し、熱中症において脳でどういった現象が起きているのか、ラット脳スライスを用いて調べています。
以下に日本法医学会で発表しました抄録を掲載させて頂きます。
In Japan, 58,729 patients with heat stroke were transported to hospitals
in 4 months (from June to September, 2013), and 88 of it died. Some of
patients have neurologic dysfunction by heat stroke, but the
pathogenesis of it remains unclear. On the other hand, the immune
system plays a crucial role in dysfunction of homeostasis. We
hypothesized that the deficiency of the immune system may cause the
neural dysfunction. Therefore, the object in this study is to elucidate the
gene expression in organotypic brain slice cultures. Adult male Wister
rats were carbon dioxide-anesthetized and quickly decapitated. After
dissection of the brain and removal of the frontal and occipital poles
(including the cerebellum), the specimens were sliced into 200 μm thick
sections. After culturing for 3 hours, the slices were incubated at 37℃ or
42℃ in a CO2 incubator for 6 hours. Thereafter, the expression of
mRNAs in the specimens was detected by quantitative real-time RT-PCR.
The treatment at 42℃ increased expression of HSP27, HSP60, GPX1,
and BCL-XL mRNA, but decreased iNOS and TNFα mRNA compared
with that at 37℃. These results show that heat stroke causes downregulation
of inflammation, suggesting that the deficiency of the immune
system may accelerate the neural dysfunction. |
